微小隐孢子虫子孢子与宿主黏附相关蛋白的筛选

Screening for relevant proteins involved in adhesion of Cryptosporidium parvum sporozoites to host cells

摘要: 利用人结肠癌细胞(Caco-2 cell)对微小隐孢子虫子孢子噬菌体展示文库进行筛选。对筛选得到的噬菌体进行克隆及测序分析,共获得5个特异的基因片段,其中1个为已知的编码微小隐孢子虫重要的侵入和黏附相关蛋白CP2,另外4个编码功能未知的蛋白。筛选得到的蛋白可能参与子孢子与宿主细胞的黏附过程。

Abstract: The Cryptosporidium parvum T7 phage display library was screened by using Caco-2 cells. Five specific gene fragments were identified by blasting sequences in GenBank, one of which encoding the CP2 protein was previously identified as a surface molecule of sporozoites and involved in parasite invasion. The others are hypothetic proteins with unknown functions. Bioinformatic analysis of these proteins indicated that they may be involved in the host-parasite interactions.
