用单克隆抗体检测疟原虫抗原的研究

摘要: 以兔抗鼠血清代替约氏疟原虫粗制抗原或恶性疟原虫作包被,采用单克隆抗体M_(26-52)的酶联免疫吸附抑制试验检测恶性疟和间日疟红内期疟原虫抗原,对69份镜检疟原虫阳性和88份镜检阴性滤纸血进行测定,以抑制率≥9%为阳性标准,结果其阳性和阴性样本符合率均超过90%,抑制率与原虫密度呈密切正相关,最低可检出2.6个疟原虫/10~4红细胞。

Abstract: This paper reports an inhibitory ELISA using monoclonal antibodies M26-32 to detect malaria parasite antigen. The characteristic of this method is to coat the polysterene microculture wells with rabbit anti-mouse IgG serum instead of P. yoelii or P. falci-parum. Examination of 69 microscopically positive and 88 negative blood samples collec-ted on filter paper revealed that, taking inhibition late≥9% as positive standard, the coincidence rate for both microscopically positive and negative samples is above 90%. The inhibition rate is closely related to parasite density, the lowest detectable level being 2.6 parasites/104 erythrocytes.
