日本血吸虫pcDNA3.1(-)/Sj20.8真核表达及其对小鼠保护性免疫作用

摘要: 为检测日本血吸虫新基因Sj20.8作为核酸疫苗能否诱导小鼠产生保护性免疫作用, 用PCR法扩增Sj20.8基因序列, 构建重组质粒pcDNA3.1(-)/Sj20.8, 肌肉注射免疫小鼠后检测发现, Sj20.8能在小鼠肌肉中短时间存在并有微弱表达, 但未能诱导小鼠产生一定水平的保护性免疫。

Abstract: Sj20.8 gene was amplified by PCR and inserted into eukaryotic expression plasmid pcDNA3.1 to construct recombinant plasmid pcDNA3.1/Sj20.8, which was then injected into the quadriceps femoris of the BALB/c mice. Results showed that the Sj20.8 antigen was low expressed in the local tissue of the mice, and was not able to significantly reduce eggs in the liver than in the control mice.
