护龈灵体外抑杀口腔毛滴虫的研究

In Vitro effect of huyinling extract on Trichomonas tenax

摘要: 以40%~80% 5个浓度的乙醇作为溶剂提取护龈灵有效成份,获取40%、50%、60%、70%和80%护龈灵醇提浸膏。实验设40%~80%护龈灵醇提浸膏组、甲硝唑对照组和空白对照组。护龈灵醇提浸膏各组分别再设5个亚浓度组,分别于125 μl对数生长期口腔毛滴虫悬液(2×105个/ml)中加入终浓度为6.25、12.5、25、50和100 mg/ml的各浓度护龈灵醇提浸膏;甲硝唑组的终浓度为10 μg/ml,每组4孔。作用12、24和48 h后,采用显微镜计数法检测护龈灵体外杀灭口腔毛滴虫的效果。并采用噻唑蓝(MTT)法检测药物作用24 h后各浓度护龈灵醇提浸膏组对口腔毛滴虫的杀灭效果。结果显示,各浓度护龈灵醇提浸膏随着药物浓度的增大或药物作用时间的延长,对口腔毛滴虫的杀灭作用增强。药物浓度为6.25和12.5 mg/ml的60%护龈灵醇提浸膏的相对杀虫率,与相同浓度的40%、50%、70%和80%护龈灵醇提浸膏之间的差异有统计学意义(P<0.01),提示护龈灵醇提浸膏在体外具有较强的杀灭口腔毛滴虫作用,其中以60%护龈灵醇提浸膏的作用最强。

Abstract: The active ingredient of Huyinling, a combination of Chinese traditional medicine, was extracted by five different ethanol concentrations(40%-80%). There were seven groups named as five Huyinling ethanol extract groups (40%, 50%, 60%, 70%, and 80%), metronidazole group and blank control. Each Huyinling ethanol extract group was further divided into five subgroups with final concentration of 6.25, 12.5, 25, 50, and 100 mg/ml, respectively. Metronidazole group was given 10 μg/ml of the drug. Each group had 4 wells with 125 μl T. tenax(2×105/ml). At 12 h, 24 h and 48 h after drug treatment, the anti-T. tenax effect of Huyinling ethanol extract was tested by microscope counting method. At 24 h the effect of Huyinling on T. tenax was examined with methyl thiazolyl tetrazolium (MTT) assay. The results showed that the higher concentration of Huyinling ethanol extract, the better effect on anti-T. tenax. 60% Huyinling ethanol extract group with concentrations of 6.25 mg/ml and 12.5 mg/ml showed higher anti-T. tenax effect than other groups (P<0.01). The ethanol extract of Huyinling granules has a remarkable effect on T. tenax, and among the groups, 60% ethanol extract shows the best anti-T. tenax activity.
