恶性疟原虫PFC0460w基因片段的克隆与生物信息学分析

摘要: 用逆转录PCR方法从恶性疟原虫3D7株红内期RNA扩增PFC0460w基因片段,克隆于pGEM-T easy载体,转化大肠埃希菌DH5α,筛选阳性克隆,PCR鉴定、测序并作序列分析。结果显示,从恶性疟原虫3D7株中扩增到3种PFC0460w片段序列,分别为618、597和543 bp,其中618 bp的片段序列与PlasmoDB数据库中的已知序列完全一致(GenBank登录号为XM_001351147)。597 bp和543 bp片段序列已登录GenBank数据库,登录号分别为JF799872和JF799873。618 bp片段编码205个氨基酸,经Robbeta服务器结构预测,其编码的蛋白质可能有5种三维结构。

Abstract: The gene fragment of PFC0460w was amplified from RNA of Plasmodium falciparum 3D7 strain with RT-PCR,and cloned into pGEM-T easy vector. The recombinant plasmid was transformed into E. coli DH5α and the positive clones were selected,which were identified by PCR and sequencing. The results showed that there were three sequences of PFC0460w fragment, respectively with length of  618,597 and 543 bp. The 618 bp fragment was completely consistent with the sequence published in PlasmoDB(GenBank No. XM_001351147),and the 597 bp and 543 bp fragments were submitted to GenBank with Accession No. of JF799872 and JF799873,respectively. 205 amino acids were encoded by the 618 bp fragment,and five kinds of protein structure were predicted by Robetta.
