日本血吸虫胆绿素还原酶的测定

摘要:   目的 探讨日本血吸虫是否含有能降解胆绿素的胆绿素还原酶(biliverdinreductase,BR)及其活性。 方法 从日本血吸虫成虫匀浆组织中提取可溶性蛋白组分,将其与胆绿素还原酶的特异性底物胆绿素共孵育 ,观察胆绿素降解情况以及时间、pH值对降解活性的影响。 结果 成虫匀浆提取物可在体外将胆绿素降解为胆红素 ,该酶活性在pH8.7时最高 ,为43.30nmol/(mg·min) ,相同pH值的不同缓冲液体系下酶活性的差异无显著性意义 ,酶促反应于15min时达到峰值。 结论 本文首次证实日本血吸虫成虫具有胆绿素还原酶 ,可降解胆绿素成胆红素

Abstract:  Objective To explore the presence of biliverdin reductase (BR) activity in adult worms of Schistosoma japonicum in vitro. Methods The soluble fraction was isolated from homogenates of adult worms of S. japonicum, and incubated with biliverdin under different pH conditions and buffers. The time dependency of this enzymatic reaction was also detected. ResultsThe soluble fraction of the homogenates of adult worms could degrade biliverdin in vitro, the BR activity was 43.30 nmol/(mg·min), with its optimal condition at pH 8.7. The rate of reaction peaked at 15 min of incubation for the BR activity. Conclusion The presence of BR activity in adult worms of S. japonicum was firstly demonstrated.
