抗杜氏利什曼原虫单克隆抗体的制备及鉴定

摘要: 本文报道七株分泌抗新疆杜氏利什曼原虫单克隆抗体(McAb)的杂交瘤细胞株的建立。这些McAb是属于小鼠IgM,IgG_1亚类。从免疫荧光的交叉反应中表现出明显的特异性,有四株McAb与其他三种利什曼原虫(热带利什曼,砂鼠利什曼,蜥蜴利什曼)以及锥虫均不起作用。另外,用活前鞭毛期加McAb作间接荧光试验,可观察到细胞膜表面的荧光反应.

Abstract: The fusion of sp2/o myeloma cells with spleen cells from mice immunized with Leishmania donovani promastigotes produced 7 hybridoma clones.In an immunodiffusion system with subclass-specific antisera, L8G5, L9E4, and L9F5 were shown to be IgG1, whereas L7B5, L7E12, L7A6 and L7C11 were IgM. The results showed that the monoclonal antibodies were not cross reactive with other species of Leishmania and Trypanosoma.Indirect immunofluorescent antibody assay with live leishmania showed that the monoclonal antibodies recognized only the antigens bound to the surface of L. donovani promastigotes.
