摘要: 应用PAGE从粉尘螨Dermatophagoides farinae浸液中分离出11条蛋白质区带。挑刺试验证明,其中7条区带的洗脱液含有过敏原,能使171例吸入型哮喘病人呈阳性反应,以6号区带洗脱液的过敏反应最强,阳性率达49.1%。全部区带洗脱液不能使所有病人都呈现阳性反应;每一过敏患者仅对其中某些区带的洗脱液出现过敏。用对流免疫电泳(CIE)证明3种区带洗脱液含有沉淀抗原,但只合少量过敏原,过敏性最强的6号区带洗脱液不含沉淀抗原。用不连续PAGE从粉尘螨浸液中分离出21种蛋白质组分,其中6种含糖蛋白,4种含脂蛋白。应用SDS-PAGE可测得40余种蛋白质组分。

Abstract: 11 protein zones were isolated from D. farinae extract by PAGE (polyacrylamide gel electrophoresis). 7 of the 11 zone extracts contained allergens, which developed positive reactions in 171 inhalant asthmatics by prick test. Among them, the 6th zone extract was the most sensitive, its positive rate being 49.1%. However, there was no extract which could produce positive reactions in all the patients, and each patient was only sensitive to some, not all of these zone extracts. The number of zones causing positive reactions and the intensity of reactions were quite different from each other. 3 of the 11 zone extracts contained precipitating antigens by CIE but very little allergens. The 6th zone extract which was the most sensitive, didn't contain any precipitating antigen. 21 components of proteins were isolated from D. farinae extract by PAGE. 6 of them contained glycoproteins and 4, lipoproteins. More than 40 components of proteins were isolated from D. farinae extract by SDS-PAGE.
