脲酶结合物ELISA测定法在日本血吸虫病诊断中应用的初步报告

UREASE-ANTIBODY CONJUGATE ELISA AND ITS APPLICATION IN THE DIAGNOSIS OF SCHISTOSOMIASIS JAPONICA-A PRELIMINARY REPORT

摘要: 本文系用脲酶结合物系统进行ELISA测定诊断血吸虫病的初步报告。自巨豆粉中制得的脲酶结晶,用戊二配合一步法与羊抗人IgG结合制成结合物。酶底物为溴甲酚紫—尿素溶液,作用后产生的氨使pH发生改变,溶液中的溴甲酚紫指示剂由黄色转变为紫色,目测观察对比分明。脲酸不存在于一般的细胞和组织中,不受内源酶的干扰,不为NaN_3所抑制,在试剂和样本中可加防腐剂易于保存。对36份血吸虫病人血清和36份正常人血清用本法进行测定,结果与用辣根过氧化物酶结合物的常规目测法一致。

Abstract: A new enzyme immunoassay system, Urease-antibody conjugate ELISA and its application in the diagnosis of schistosomiasis japonica was reported. Urease was extracted and crystallized from jack bean meal. Urease-conjugate was prepared by the one-step glutaral-dehyde method. The substrate solution was made up with bromocresol purple and urea. Urease catalyzes the hydrolysis of urea to NH3, and pH shift causes a vivid colour change from yellowish to purple. This enzyme-substrate system offers a number of advantages, the substrate is stable, easy to obtain and without harm to human beings, and the colour change between negative and positive reaction is unmistakable. As urease is unaffected by the presence of the preservative sodium azide in reagents, the substrate may be prepared and stored until use. As urease does not exist in mammalian tissues, no endoge-neous activity occurs when urease-antibody conjugates are used to detect antibodies. A total of 36 normal persons and 36 schistosomiasis patients were tested by ELISA using urease-conjugate, the preliminary results showed that both sensitivity and specificity are promising.
